Detection of Cymbidium Mosaic Uirus and Odontoglossum Ringspot Virus in Orchid Plants by Tissue-blot Immunoassay

Abstract

|
The application of tissue-blot immunoassay (TBIA) for detecting Cymbidium mosaic virus (CyMV) and Odontoglossum ringspot virus (ORSV) from infected cymbidium and cattleya plants was evaluated. CyMV and ORSV were readily detected in tissue blots made from leaves and roots of infected cymbidium and cattleya by both direct and indirect TBIA. Tissue-blots made on PVDF (polyvinylidene difluoride) membrane gave much clearer specific color reaction than those made on nitrocellulose, or nylon membrane. Non-specific reactions were completely eliminated when the CyMV or ORSV rabbit polyclonal antibodies, or alkaline phosphatase-conjugated CyMV or ORSV polyclonal antibodies were absorbed with sap from non-infected host plants from which the respective viruses were purified. Root tissues were more efficient than leaf for detecting CyMV and ORSV by TBIA.
